I always did parking brake on, car in neutral when the car was being transported (in addition to very secure tie downs and wheel chocks); probably not a big risk, but some risk of damaging the transmission from the car rocking back and forth slightly with the car in gear.
I'd recommend that. I should also mention that some of the competition car guys will use a set of Porsche Motorsport Center Lock Tie Down items, only applicable to center-bolt cars, obviously. See the pair of pre-production 918s on the freeway in Southern California. The trouble with these is you have to get them via Porsche Motorsports (or at least that used to be the case), and then you have the hassle of swapping center-bolts.
