In the case of the OP, the offending ECU is likely the one causing the misfires.
The return plumbs into the large pipe at the front of the car and all the coolant gets back to the block via that big hose.
Not true. Each ECU will monitor the four cylinders for that bank. Once a misfire is detected in bank 2, it reports the misfire to the Master ECU...
If it goes with the ECU, it's the ECU faults. When installed, the ECUs function as Master (B1) or Slave (B2), but you can swap it left to right...

Did you look at the end of the 16mm hose that feeds coolant to the heater valves? I bet it looks like that.
