Yesterday I ran over a screw (with washer on it) in my '06 Denali. I removed the screw and prepared to plug the hole when to my surprise it didn't puncture the tire. It went straight in over the meat of the tread. My question is, do you think the tire is compromised at all? I half considered swapping the spare.

On a normal tire, I would say change it. But since it's an 06 Denali, it should be ok.

If it just went into the tread, youre fine. There will come a day when all that "meaty" tread won't exist anymore due to wear.

Youre lucky. I walked out on a Sunday morning about 4 weeks ago to a flat tire. Hadda break out the plug kit! Been a long time since I did a plug. Thank God its like riding a bike.

I was lucky it went into the meat of the tread. I had a plug kit and was all ready to clean it up and put a plug in. I think I got lucky this time. I am planning on taking the car to Montauk next weekend and didn't want any surprises along the way.

Doesn't a plug screw up the belts? I've been told to only patch a tire from the inside.

Ive driven plugged tires for tens of thousands of miles and never had an issue. It depends where the puncture is. Some spots on a tire should be patched and not plugged. NEVER repair a puncture to the side wall. NEW TIRE ONLY!

    False.

    True.

If it only went in half way, or partially through the tire, there will be a gap in the belts. This will cause a bubble and will eventually cause the tire to separate from the belts.

My advice, get it plugged and patched. Yes, both. You must ask them to push it all the way through, plug it, then patch it on the inside.
